CD-349 has a vasodilatory actionappearing more slowly than nicardipine when administered intravenously. The binding rate to the L-type calcium channels for CD-349 is much higher tha that for nitrendipine in the crude membrane preparation, whereas the uptake of CD-349 into tissue was somewhat slower tha that of nitrendipine in bovine vessels. The results of the present study showed that the distribution intu tissues and the release from the tissues occurred more slowly with CD-349 than with nicardipine in cardiac, vascular and skeletal muscular tissues. In addition, the present study revealed tha CD-349 and nitrendipine inhibited the L-type Ca current in single ventricular myocytes in a similar manner with time. The results of the tissue distribution and release study explained well the time to reach the maximal effects of CD-349 and nicardipine, and the present electropharmacological study demonstrated that the time of onset to exert Ca antagonistic actions of dihydropyridine compounds is completely dependent on the rate of drug diffusion into tissues but not the channel binfing rate even in isolated single cells.
