| 内容記述 | Hypertension, diabetes, and dyslipidemia are lifestyle-related diseases caused by longstanding lifestyle habits. In addition, when these diseases coexist, the intake of multiple medications is necessary, which increases the potential for drug interactions. We have already analyzed and quantitatively elucidated the relationship between the pharmacokinetics and hypoglycemic effects of antidiabetic glimepiride. This study focuses on the hypoglycemic effects of glimepiride and the antilipidemic bezafibrate in combination, and conducts basic investigations on their pharmacokinetics and effects in rats. Regarding the pharmacokinetic characteristics of bezafibrate, it was found that, its terminal half-life (t1/2) is longer, its total body clearance (CLtot) is lower, and its mean residence time (MRT) is longer, indicating prolonged retention in the body compared with glimepiride. Additionally, when both drugs were co-administered, the pharmacokinetics of glimepiride remained largely unchanged despite an extension in its t1/2. When bezafibrate was administered separately, it exhibited a similar degree of hypoglycemic effect as the control group and lasted approximately 60-120 minutes after administration. In the case of co-administration, the hypoglycemic effect of glimepiride was almost the same as that of glimepiride separately. This result possibly reflects the hypoglycemic effect of the co-administration. Although no significant fluctuations were observed in the hypoglycemic effect when both drugs were used concurrently, co-administration led to an increase in the tissue distribution of bezafibrate. This suggests that the long-term efficacy of bezafibrate may have been affected. As this effect takes approximately one week to manifest, unlike the hypoglycemic effect, it will be necessary to consider it in future long-term plans. |
